Each review is written from my personal point of view and on this occasion it seems that I need to make a correction.The influence of in vivo cytokine treatment on antigen presentation by dendritic cells is dependent on the dendritic cell maturation stage.
In the course of an infection with microbial pathogens, dendritic cells (DCs) are activated to present pathogen-derived antigens in an MHC-restricted manner. While it is well accepted that DCs are important for the initiation of the immune response, there is no consensus on their role in the regulation of this response. Here we studied the influence of in vivo injection of cytokines on the presentation of exogenous Ags by murine DCs. Bone marrow-derived DCs were generated in vitro, pulsed with ovalbumin and injected into the footpad of mice. Mice were treated with recombinant murine IL-10, IL-12 or IFN-gamma on day 0 and day 7 after DC inoculation, and Ag presentation by the injected DCs was measured in footpad lymph node cells on day 9. The IL-10-treated mice had reduced Ag-specific proliferative responses compared to untreated mice, while IL-12 treatment resulted in Ag-specific proliferative responses similar to those of untreated animals. IL-10- and IL-12-treated mice also had less Ag-specific IFN-gamma production. In contrast, mice treated with IFN-gamma had IFN-gamma responses similar to those of untreated mice. Analysis of cell surface markers suggested that DC maturation stage is critical in the response to cytokine treatment. Only IFN-gamma-treated DCs of the Langerhans cell type were able to process and present Ag, while DCs generated in vitro from bone marrow cells failed to do so. These results demonstrate that cytokines can have profound effects on the functional phenotype of DCs and suggest that the effect of cytokines on immune responses is dependent on the DC maturation stage.On the occasion of the International Women’s Day, I’d like to honor the women who have mentored me in my writing career and the many women (and a few men) who have helped me create and build this website.
